Are you on a Mac or Linux computer? Do you seem the same thing for all files on the Desktop regardless of location? All files in that particular folder on your desktop?
Also, is this with vanilla Sublime or do you have any third party packages also installed?
The EPERM error code (which is what Operation not permitted equates to) generally relates to your trying to carry out an action without the appropriate privilege. That’s different from EACCES (Permission Denied) in that it (in my experience) generally denotes that you’re doing something that requires higher privileges than you currently have.
Off the top of my head I’m not entirely sure what could be causing that for a save operation on a file that you (presumably) successfully loaded previously.
